Part I: Direct observation of nucleosome in DNA frame using HS-AFM
The nucleosome is one of the most fundamental units involved in gene
expression and consequent cell development, differentiation, and expression
of cell functions. We report a method to place reconstituted nucleosomes
into a DNA origami frame for direct and real-time observation using high-
speed atomic-force microscopy. Multiple nucleosomes can be placed in one
frame, providing a promising platform to study nucleosome interactions and
chromatin structure.
Part II: Retrieval of membrane proteins during vacuolar/endosomal
transport in plant cells
The distinctive protein and lipid composition underlies the distinct function
of each organelle, regulated by balanced anterograde and retrograde
membrane trafficking. Although the vacuole is pivotal in various plant
functions, the existence of retrograde transport from the plant vacuole
remained unverified. We identified the machinery to retrieve the plant
specific SNARE VAMP727 from vacuolar membrane in Arabidopsis.
In addition, I would like to introduce my future plan about the involvement
of vacuolar SNARE and plant floral development.- 問合せ先
