E. coli is able to survive exposure to surprisingly low pH, through the expression of a number of acid resistance systems. The regulation of these systems and the ways in which they depend upon growth stage is complex
and not fully understood. We are using a combination of high throughput measurements of multiple promoter activities over time in a range of mutant backgrounds, laboratory-based evolution of novel acid-resistance phenotypes, whole genome sequencing, and network inference from microarray data, to improve our understanding of this complex response. In this seminar I will present some of our recent data in this area, which gives us new insights into the network of regulators that control the response to low pH, and into the
ways in which E. coli detects acid.
